Isshiki, not Jura, will go down as Boruto's best-written villain
Boruto is a series that doesn't get a lot of credit when it comes to its villains but Jura has recently been praised in the Two Blue Vortex time-skip because of his demeanor and connection with the Ten-Tails. However, while he is an antagonist with some truly compelling elements and narrative, the best-written villain in the series is Otsutsuki Isshiki.
The former leader of Kara has proven to be one of the biggest threats in Boruto and also someone who has influenced certain events in the story, which are the marks of a great villain.
Moreover, his personality is displayed in a subtle way, adding to his characterization and making him a believable and efficient threat to the main cast.

Explaining why Isshiki is the best-written villain in Boruto and superior to Jura in a lot of ways
One trait that has defined Otsutsuki Isshiki throughout his run in the series is his ability to stay calm at most times, as evidenced early on when he was informed that the "vessel" Kawaki had fled.
Instead of lashing out or losing patience, he takes this as an opportunity to research Kara to determine who the real traitor is in his organization.
There was also when he talked with Delta as he was eating and the former was lashing out and getting visibly angry but Isshiki stayed calm, which showed him in a position of power.
When Kashin Koji wants to head to the Hidden Leaf Village, the Otsutsuki doesn't restrict his subordinate and allows him to go there, highlighting his intellect since he views this as an opportunity to analyze who is the traitor.
On the other hand, while Jura has managed to be a compelling villain and is deeply connected to the lore of the series, his intelligence and ability to deal with obstacles are yet to be seen.
Therefore, Isshiki proves to be a capable leader and also someone who can remain calm in times of conflict, which was quite useful for him during his run in the series.

When dealing with Amado's betrayal, Isshiki didn't react out of emotions or conflict but rather through planning, as evidenced by how he got the better of Kashin Koji in the Boruto manga.
This is something that makes him stand out from most Otsutsuki in the series in the sense that he has a deep understanding of human nature, which is an element that is a direct contrast with Jura.
While Jura has been constructed as someone who is trying to understand people, Isshiki has managed to get a certain perspective of humanity and see what makes them tick.
This shows a degree of willingness to learn, which is a direct contrast to most Otsutsuki logic. This is shown by how he apologizes to Naruto Uzumaki for using shoes in his home, which is displayed as a bit of dark humor.
All of this comes to fruition when he is outmatched by Kawaki and reaches his end in the Boruto manga, which sees him breaking down.
This makes his aura of calmness and intelligence fall apart, making his defeat all the more enjoyable to witness.

Isshiki is the best villain in Boruto because of his charisma, how he influences the plot, and his intelligence. He proved to be different to the Otsutsuki in general and also posed a major threat to the main cast.
