After his high school career, he committed to playing college football at James Madison University for the James Madison Dukes. His collegiate journey began in 2020 at James Madison, where, as a freshman, he made an immediate impact, featuring in all eight games with four starts. He led the team with 33 receptions, accumulating 603 yards and contributing six touchdowns. In his sophomore year, 2021, Wells Jr. continued his stellar performance, starting all 14 games and setting a school record with 83 receptions, amassing an impressive 1,250 yards, and securing 15 touchdowns.

Antwane Wells Jr. decided to transfer to the University of South Carolina and play for the South Carolina Gamecocks in 2022. He seamlessly became a starter in his inaugural year with the team. Currently donning the No. 3 jersey for the Gamecocks, he has solidified his position as a standout wide receiver. In recognition of his outstanding contributions, he earned the prestigious title of First-team All-SEC in 2022, highlighting his prowess on the football field. Antwane Wells Jr. transferred to the University of Mississippi and will play for the Ole Miss Rebels in the 2024 season.
