The Baltimore Ravens selected him as the 16th overall pick in the first round of the 2017 NFL Draft. Since joining the Ravens, he has become a key player on their defense, earning a first-team All-Pro honor in 2019 and Pro Bowl selection in 2019, 2020, and 2022. As of Week 117 in the 2024 NFL season, he has recorded 436 total tackles, 6 sacks, 15 forced fumbles, 5 fumble recoveries, 90 pass deflections, 19 interceptions, and three defensive touchdowns.

He was born on July 8, 1996, and raised in Hoover, Alabama, Humphrey was a standout athlete at Hoover High School, where he excelled in both football and track. He was named to the USA Today All-USA high school football team in 2012 and 2013 and was a three-time All-State selection. Humphrey attended the University of Alabama, where he played college football for the Crimson Tide in 2015 and 2016. Humphrey helped Alamaba to a CFP National Championship in 2015.
